Flame Hashira Kyojuro Rengoku receives new orders: Travel to the Mugen Train, where over forty people have gone missing, and conduct an investigation. Leaving the Demon Slayer Corps Headquarters, he sets off on this new mission.

Tanjiro's group, fresh from completing their training at the Butterfly Mansion, head to their next mission, the Mugen Train. There they rendezvous with Rengoku, who arrived ahead of them. Tanjiro is overwhelmed by the always-energetic Rengoku, Zenitsu is frightened by the prospect of facing demons as usual, and Inosuke is immensely excited by the sight of the huge machine. Tanjiro, Nezuko, Zenitsu and Inosuke join Rengoku on the Mugen Train. Having heard that there may be demons on the train, they are on high alert, but they find themselves falling into a slumber. Nezuko, Zenitsu and Rengoku fight against Enmu to protect the passengers. Meanwhile, Tanjiro and Inosuke discover the location of Enmu's neck. They work in tandem to thwart Enmu's Blood Demon Art and sever Enmu's neck. The Mugen Train writhes in pain and derails. What will be the fate of the 200 or so passengers as well as Tanjiro and his friends?

Though Uzui's three wives have been infiltrating the Entertainment District to gather information about demons, Uzui has lost contact with them. In order to find out their whereabouts, Tanjiro and his comrades begin their infiltration...

To gather information, Tanjiro, Zenitsu and Inosuke have infiltrated each of the Houses where Uzui's wives are working. Rumors about Makio make Inosuke suspicious, and he charges into her room. Meanwhile, hearing a young girl crying, Zenitsu rushes to the source of the sobs, only to find the distressed girl in a room in shocking disarray. Behind him, a shadow creeps ever closer...

After his encounter with Upper Six, Daki, Zenitsu is sure that she is a demon; however, Daki also realizes that he is a member of the Demon Slayer Corps. Shortly after, Zenitsu disappears. Tanjiro and Inosuke, who have gathered for their regular meeting, are notified of Zenitsu's disappearance by Uzui and told to leave the Entertainment District.

As Nezuko rushes to aid Tanjiro during a critical moment, her appearance begins to gradually take on a demon form as she sustains Daki's attacks. Nezuko is able to overpower Daki as her demon form accelerates; however, she turns her attention to innocent bystanders injured during the battle and charges towards them. While Tanjiro attempts to restrain Nezuko, Daki draws near...

Uzui battles furiously, taking on both Daki and the other demon who emerged from her body, Gyutaro. But he's gradually losing strength due to the poison from Gyutaro's scythe. Just then, Inosuke, the sleeping Zenitsu, and Tanjiro, who's managed to get Nezuko to sleep, all come rushing to Uzui's aid...

Hinatsuru's use of wisteria poison leaves Gyutaro momentarily vulnerable, but Tanjiro and Uzui's attacks fall just short. Using his Blood Demon Art, Gyutaro vanishes from Uzui's sight only to reappear assaulting Hinatsuru. Tanjiro and Uzui attempt to save her, but they are thwarted by Daki's Belt.

Amid the wreckage from the buildings destroyed by the destructive attacks of Daki's Belt, Tanjiro awakens. Zenitsu is trapped under the rubble and Inosuke has been impaled by Gyutaro's scythe, while Uzui has lost an arm and his heart has stopped. As Gyutaro taunts Tanjiro by calling him a disgrace, all seems hopeless until...

As the fierce battle draws to an end, Tanjiro and Uzui behead Gyutaro just as Zenitsu and Inosuke behead Daki. But then, Gyutaro unleashes a devastating Blood Demon Art, burying the city in rubble. When Tanjiro awakens, he joins Nezuko and Zenitsu, only to find Inosuke and Uzui on the brink of death from Gyutaro's poison...
